Catalent & TFF Pharmaceuticals Announce New Inhalation Dry Powder Development & Manufacturing Agreement


Catalent and TFF Pharmaceuticals, Inc. recently announced their collaboration agreement focused on the generation, testing, and manufacture of dry powder formulations for a range of biotherapeutics through the application of TFF Pharmaceuticals’ patented Thin Film Freezing (TFF) technology. Under the agreement, Catalent will provide its scale-up expertise and manufacturing capabilities to TFF Pharmaceuticals as its preferred development and manufacturing partner, and Catalent will be able to offer its customers access to the innovative TFF technology, which has the potential to increase the adoption of the pulmonary route for the systemic delivery of biotherapeutics.

Thin Film Freezing may potentially be used to generate dry powder formulations for inhalation, and it complements Catalent’s existing capabilities in spray drying for the development of inhalable drugs established by its 2021 acquisition of the manufacturing and packaging operations of Acorda Therapeutics in Boston, Massachusetts.

TFF Pharmaceuticals’ Thin Film Freezing (TFF) platform was designed to improve the solubility and absorption of poorly water-soluble drugs and is particularly suited to generate dry powder particles with properties targeted for inhalation delivery, especially to the deep lung, an area of extreme interest in respiratory medicine. The TFF process results in a “Brittle Matrix Particle,” which possesses low bulk density, high surface area, and typically an amorphous morphology, allowing the particles to supersaturate when contacting the target site, such as lung tissue. Based upon laboratory experiments the aerodynamic properties of the particles are such that the portion of a drug deposited to the deep lung has the potential to reach as high as 75%.
